A sensor for the continuous monitoring of pH in sea water, having the configuration of a probe, is described. The characteristics of this probe, which uses phenol red adsorbed on Amberlite resin as a chemichromatic element on top of an optical fibre, are reported and the effects of interfering parameters (variable salinity and temperature of sea water) are assessed. The possibility of enhancing the working range is discussed, and a procedure for rendering the range suitable for marine monitoring is presented. 相似文献

In order to obtain OH concentration measurements in atmospheric pressure flames from saturated fluorescence signal, we have developed a stationary four-level model. A general relationship between the measured upper state population and the total number density has been derived. The effects of the imbalance between the rotational transfer rates in the upper and ground states is analyzed. We show that in the case of comparable rotational rates, the results of our treatment are similar to the ones obtained with the balanced cross rate model. A set of complementary data which should facilitate absolute OH concentration measurements is also pointed out. 相似文献

The cycloaddition reactions of ‘all-carbon’ 1,3-diazabuta-1,3-dienes with a few conjugated and unconjugated alkynyl ketenes are described. Several methods have been described in the literature for removal of DNA from protein samples prior to proteome analysis. They in general involve protein precipitation techniques. In other protocols, DNAse treatment is suggested prior to precipitation of proteins in excess acetone. All these methods have been evaluated and found to perform poorly in DNA removal, as illustrated by two-dimensional (2D) maps where horizontal and vertical sample streaking are still substantial. Such removal is in general necessary in tissue lysates and especially when analysing sub-cellular organelles, such as nuclei, where the high DNA levels strongly interfere with proteome analysis. Another method is proposed here for efficient DNA removal: two-phase extraction of DNA in chloroform/phenol/isoamyl alcohol, a procedure commonly used to rid DNA samples of protein contaminants, but rarely applied to protein preparation. This extraction is not very efficient if performed at slightly acidic to neutral pH values, but it performs extremely well at pH values of 9.5 or higher. The 2D maps thus obtained of Escherichia coli lysates as well as extracts from purified nuclei of eukaryotic cells are not only devoid of any vertical or horizontal streaking, but exhibit many more spots, especially in the alkaline region of the 2D gels, suggesting that these basic proteins were in general lost to proteome analysis due to co-precipitation in tenacious protein–DNA complexes. It is hypothesized that the alkaline pH values adopted in the two-phase extraction help to fully disrupt any residual DNA–protein complexes, due to strong Coulombic repulsion. 相似文献

Polysiloxane-modified hybrid Nafion membranes were prepared by casting a mixture of Nafion solution and a precursor of acid functionalised polysiloxane based on tetraethoxysilane and a mercaptan-organoalkoxysilane.Scanning Electron Microscopy (SEM) and Atomic Force Microscopy analysis revealed that the functionalised polysiloxane was dispersed either as finely nanosized inclusions or as coarse domains depending on the rate of the solvent evaporation during the casting procedure. In particular the slower is the rate of solvent evaporation the more interpenetrated and homogenously dispersed at nanosized level is the polysiloxane inside the Nafion membrane.The hybridization process increases the thermal stability of the membranes of about 50 °C relatively to the unmodified Nafion. Small angle X-ray scattering (SAXS) analysis reveals that the hybrid membranes exhibited the typical morphology of Nafion consisting of distinct hydrophilic and hydrophobic domains.Water vapor sorption and proton conductivity were measured varying the temperature (up to 120 °C) and the water activity conditions (from 0.1 to 0.8). The polysiloxane network always increases the water vapor uptake of the membranes and increases significantly the proton conductivity at higher temperature depending on the type of morphology developed by the manufacturing method. In particular hybrid membranes exhibiting nanosized polysiloxane dispersion show a proton conductivity which is up to one-and-half time higher than Nafion recast membrane at high temperature and low water content. 相似文献
